Cellular uptake of polymeric nanoparticles by bovine cumulus-oocyte complexes and their effect on in vitro developmental competence

چکیده

Polymeric nanoparticles (NPs) are produced using bio-compatible and bio-degradable materials such as PLGA (Poly(lactic-co-glycolic acid)). This technology provides a valuable tool to deliver molecules the subcellular level with relatively low risk of cytotoxicity. However their use in field reproductive biotechnology is not yet scientifically substantiated. The aim present study was test if NPs can be taken-up by cumulus-enclosed oocytes first step towards potential oocyte-targeted applications enhance oocyte quality fertility. We conducted series experiments bovine vitro maturation model FITC-conjugated internalization (using laser-scanning confocal microscopy) effect some important physical (particle size) chemical (conjugation PEG) modifications. show evidence that cumulus cells less extent enclosed regardless NP size. transfer appear transcellular (via transzonal projections) paracellular zona pellucida). were detected vicinity quick 2 h post-exposure protein-free medium did compromise cell viability nor subsequent early embryo development or quality. These results suggest may have promising carriers for drug molecule delivery targeting oocytes.

the effect of e-64 on the developmental competence of sheep cocs during in vitro maturation

in the present study, the effect of e-64 at different concentrations (0.5, 1, 5 and 10 µm) added to (1) the ivm medium on oocyte nuclear maturation and developmental competence of ovine oocytes, and (2) to the ivc medium on embryonic development of ovine embryos were investigated.

P-110: Effect of Increasing Amount of Oocyte Secreted Factors on Cumulus Expansion of Bovine Cumulus-Oocyte Complexes

Background: In vitro maturation is a good method to decrease cancer risk of superovulation by gonadotropin hormones. A paracrine effect of oocyte secretions on oocyte developmental competence is under investigation. Apart from oocyte maturation, ovulation in vivo requires a precise control of extracellular matrix modification. Cumulus cells secrete hyaluronan to form a muco-elastic extracellula...

Effect of Different Harvesting Techniques on the Recovery and Quality of Bovine Cumulus Oocyte Complexes

This study investigated the effect of different harvesting techniques on the recovery rate and quality of bovine cumulus oocyte complexes (COCs) and their subsequent developmental competence including in vitro maturation, fertilization and blastocyst formation. The COCs were retrieved from slaughter ovaries using aspiration, slicing, or aspiration followed by slicing. The COCs were examined mic...
